DNA polymerase of reticuloendotheliosis virus: inability to detect endogenous RNA-directed DNA synthesis.

Purified preparations of reticuloendotheliosis virus (REV) appear to lack the endogenous RNase-sensitive DNA polymerase activity present in most, if not all, avian RNA tumor viruses. Although no endogenous RNA-directed DNA polymerase could be detected in REV, we have been able to demonstrate the presence of a virion-associated DNA polymerase by employing exogenous synthetic homopolymers as temp...

RNA-directed DNA polymerase and virus-induced leukemia in mice.

Rifamycin antibiotics, which inhibit RNA-directed DNA polymerase of Rauscher leuckemia virus, prevent the leukemogenic activity of the virus, and the effect of leukemogenesis correlates with the magnitude of inhibition of the purified enzyme. This inhibition of enzyme activity by rifamycin SV derivatives is due to a relatively tight binding between the enzymes and the inhibitors, yet the bindin...

Primer recognition by avian myeloblastosis virus RNA-directed DNA polymerase.

Tryptophanyl-tRNA was specifically labeled at the 3' end with [3H]tryptophan and cleaved in half with RNase under denaturing conditions, and the 3' half was shown to hybridize exclusively at the 5' end of avian myeloblastosis virus RNA. The RNA-dependent DNA polymerase of avian myeloblastosis virus is capable of efficiently binding the 3' half of the primer molecule.

Stimulation of Rauscher leukemia virus DNA polymerase DNA-directed DNA synthesis by cationic trypanocides and polyamines.

Activated DNA-directed DNA synthesis catalyzed by Rauscher leukemia virus (RLV) and other type C mammalian retroviral DNA polymerases is uniquely stimulated by biologically active polyamines. Cationic trypanocides may act as antagonists of polyamine function. As described here, several cationic trypanocides stimulate RLV polymerase-catalyzed DNA-directed DNA synthesis at concentrations signific...

Hamster leukemia virus: lack of endogenous DNA synthesis and unique structure of its DNA polymerase.

Infectious hamster leukemia virus (HaLV) contains a DNA polymerase different from those of murine and avian viruses. No endogenous reaction directed by the 60 to 70S RNA of HaLV could be demonstrated in detergenttreated HaLV virions, nor could the purified DNA polymerase copy added viral RNA. The virion RNA could, however, act as template for added avian myeloblastosis virus DNA polymerase and ...
